Developing and producing cell and gene therapies, as well as mRNA vaccines, requires assessing sample critical quality attributes (CQAs). This includes analyzing vector packaging – the proportion of vectors carrying the therapeutic DNA or RNA cargo – as well as measuring titer and sample purity.
Ideally, vector analytics would be run quickly on little sample, so that production and purification processes can be monitored frequently. The ability to gain insights throughout processing facilitates early identification and correction of faults, speeding up optimization. However, most analytical methods in use today are slow and/or sample-hungry, and cannot be relied on to deliver rapid, in-process information.
Mass photometry and related macro mass photometry, on the other hand, are two technologies offering fast, powerful vector analytics capabilities. Returning results in minutes and using little sample, both technologies offer a way to easily track key attributes during vector production and purification. Based on single-particle analysis, mass photometry technologies also offer exquisite sensitivity, enabling the detection and quantification of distinct subpopulations within samples.
Mass photometry is ideal for use during adeno-associated virus (AAV) production. It can measure not just the AAV empty/full ratio, but also quantify partially filled and overfilled capsids, among other sample attributes. With software that enables use in GMP-compliant settings, mass photometry is also suitable for use in viral vector manufacturing.
Macro mass photometry, meanwhile, can be used to assess the purity and stability of vectors such as adenovirus and lentivirus. It can also, for example, analyze adenovirus packaging, and its unique multiparametric readout correlates with lentivirus physical titer.
With each measurement taking just a few minutes, requiring very little sample and minimal training – mass photometry technologies put the power of single-particle vector analytics within easy reach.
