This session will highlight new research focused on optimizing AAV6 upstream production – an essential step in improving the safety and affordability of gene therapies. Using a design of experiment (DoE) approach, the Sartorius BIA Separations team systematically identified the optimal conditions to achieve a high full capsid ratio. Further optimization was conducted with the Ambr15 system, while CIM SO3 monolithic plates enabled efficient high-throughput purification of upstream samples. Mass photometry was used to accurately assess capsid species ratios, supporting more precise and scalable production.
