Mass photometry is an advanced analytical technique. It measures the interference between light scattered by individual biomolecules in a sample and the light reflected by the measurement surface. The resulting signal, known as contrast, directly correlates with mass, allowing for precise mass calculations. To ensure accuracy, molecules of known mass, called calibrants, serve as reference points. MassFerence P2 provides a single standard protein species of 3640 kDa, enabling straightforward and reliable mass conversion. Given that the contrast-to-mass relationship remains linear within the relevant range, a single reference point ensures accurate calibration. MassFerence P2 is supplied in a dry form, with each box containing three vials, sufficient for 10 calibrations per vial once reconstituted. It is easy to prepare, requiring only simple dilution steps before use. The use of Massference P2 enables faster workflows for data analysis in DiscoverMP software.

Does MassFerence P2 have buffer restrictions?

MassFerence P2 has been tested in various buffers commonly used in AAV purification processes, where the calibrant consistently provided accurate results. However, due to its nature, we cannot exclude the possibility that MassFerence P2 might be unstable in some buffers. As such, detergents should be avoided, as they significantly impact the product.

Can I aliquot and store/freeze MFP2 after resuspension?

No, this will impact the product’s stability. MassFerence P2 remains stable 5 days after resuspension at 4°C due to its nature. Other storage conditions might affect the calibrant quality and reduce the calibration accuracy.

Can I use MassFerence P2 to calibrate AAVs on my TwoMP system?

No, Refeyn does not recommend using MassFerence P2 on a TwoMP system. The nature of the MassFerence P2 calibrant makes it more visible in the low mass range, which can bias the results when using the TwoMP system, and the performance would not be guaranteed. Therefore, we do not recommend it for our TwoMP system.

How reliable is MassFerence P2 as a one-point calibrant compared to multi-point calibrations?

MassFerence P2 can be reliably used as a one-point calibrant. Testing at Refeyn has shown no significant difference between one-point, two-point, or three-point calibrations. All three methods demonstrated a linear contrast-mass relationship and produced highly comparable results.
